Using the TKG.io online tool, how can I define my own macros? (I have a RedScarfII+)

Currently TKG doesn't support user defined macro feature. I have a plan to support this feature in the future, but it's hard to support it with current configuration construction.

You have to clone the source code of the keyboard (e.g. for RedScarfII+ the source code is here) and build your own firmware with your macros defined in it at this moment.
