When you edit a translation ini file using your extension, will this work as an override?
That means, when you update extensions or languages in Joomla, will all edits within the Localise extension be destroyed and undone? How to make changes permanent, so that they never will be destroyed by software updates?

No.
Localise (or core) let's create overrides.
If one modifies an ini file which is part of a pack, next time the pack will be updated, the changes will be lost.
