Does Multi_Hit include multiple variants at the same position?

Hello, I was wondering if multiple variants found in the same locus of a gene would be considered Multi_Hit. Or would these only be counted as one hit and that additional mutations will have to occur on different positions of the gene to be considered Multi_Hit? Thank you!
